Specimen preparation for Scanning Electron Microscopy

The Scanning Electron Microscopy (SEM) is a method for ultrastructural analysis of biological surface morphology. Samples prepared for SEM analysis are made conductive with a few nanometre gold coating (sputtering). When they interact with the electron beam, the gold coating gives back a secondary electron signal producing an high resolution image of the three dimensional sample structure.
