Macromolecules (spin labeling)

EPR spectroscopy allows to study the structure, function and dynamics of macromolecules and cellular components, the intramolecular distances and the specific role of some amino acid residues through site-specific mutagenesis in absence and presence of perturbator agents.

Since the mobility of spin labels depends strictly on the macromolecular structure, any alteration of this structure, induced by the microenvironment, has effects on the EPR signals generated by the spin labels.

For this purpose, the spin labeling technique is used, which consists in marking specific amino acid residues of proteins with appropriate paramagnetic molecules (spin labels) through covalent bonds.
